What script? That option merely changes how selecting objects/components in the viewer with the mouse affects the selection.

Let me guess…perhaps the question is:
Is it possible to control selection behavior via Python script.
Is it possible to call selectGeometry(…) with “Remove from selection”
activated without worrying about pressing Ctrl key

I expect the answer to both those questions is currently no, but I'm not 100% sure.
